Does all the blank cells have to be filled in?

The general rules do not require this, but for the IB and competition puzzles, all blank cells need to be filled in.

In 10. LONG DIVISION what does it mean ONE on the top? Is that the result of division?

the long division is in the format NUMBER = DIVISOR*DIVIDEND_PLUS_REMAINDER
in the example EIGHT is the NUMBER, SIX is the DIVISOR, ONE is the DIVIDEND(result) and T is the REMAINDER.

Placing the words in the grid is sufficient for receiving full credit, marking the clues is not necessary.

The clue number at the first starting letter will be same (common) in this case. Like how a regular crossword puzzle is numbered.
